There is a vacancy on my Parish Council or which I am the Chair. As a councillor, am I allowed to take part in a vote to fill the position by co-option if my wife is one of the applicants?

Depends what it says in your standing orders on co-option procedures. Of course it would fulfil the requirement of councillors for integrity by choosing not to take part in the voting and explaining why not, to be recorded in the minutes.
